I used to recommend Adaware, then it was Spyware Nuker (I still love the guys that wrote that software), but today, I can't recommend anything other that Microsoft Windows Defender.
Microsoft released this software about a year ago and called it the Anti-Spyware Beta. It has recently changed names to Windows Defender. It's free, although I've heard rumors that they might start charging for it at some point.

It's in it's 2nd beta and is rock solid stable. It works very well. I haven't had a spyware infection problem since I've been running it.

Spybot is okay, but the other answer is better. Lava soft Ad-Aware SE is the best and not only that, it's free. You can download a version that will prevent allow you to control whether spyware stays on your computer or not and a simple, inexpensive upgrade gives you some simple, but incredibly effective tools. I have it on my desktop and scan almost everyday while I do other things.

Use multiple (anti-)spyware programs (Spybot and Ad-aware).

they are both Good Programs and are both free but they have both let me down lately i now use a program called SuperAntiSpyware as it saved me from having to do a format after a really bad infection. PS i used to sware by SpyBot but SuperAntiSpyware superseeds it :)
