I don't know if it's the best, but I prefer Sony Sound Forge. It's a great application and is easy to learn and use. However, the features of it are practically limitless. Between that and Sony Acid Pro, I can pretty much create any track or mix I want.

Another great software, though used mainly for creating loops and audio is a program called Fruity Loops. I've used it only for a short bit, however it hasn't been enough to be able to give a thorough description on it.
