The best way to to fly and you should wait until you are in bangkok to book a flight because it will be cheaper. It might cost about 100usd. If you don't have the money it should cost you about10 bucks to take a bus there. You will have to swich from bus to truck at the border. The roads are unbarable trust me. I still have a pain in my knee (being cramped in the back of a ranger)from it three years ago. It is an experience to say the least and fun to talk about it. And I got some amazing pictures.
Yes, if you do take the bus they will take you directly to a hostel that they already have lined up. They will tell you that they don't but they do. It happens all over SEAsia. And they get some money for doing it. All you have to do is walk to a different guesthouse or get a motorbike to take you to a different one if you want. I never stayed at the guesthouses that I was dropped off at. Also at the border they make you wait awhile so that you will spend money at that little resturant (they take a cut of money from that also). But remember these people are very poor. If someone offers to carry your backpack across the border (there is a short walk) let them and give them a dollar they need it. At least they are not begging for it.
If you want a good cheap place to stay I recommend the Mahoganny Guesthouse. It is one of the older ones but it is very nice and the owners will help you with anything you need. They also have a patio type thing in the front with couches where all the guest hang out at night. It was so fun! Only do the temples for three days because by the third you are going to be so tired! I could barely walk anymore! Try the tranchulas if you get offered a fried one.
I recommend going to Phenm Pehn if you have a few extra days. I took the boat and that was also amazing the views were breath taking. And there is several really cool things to see there. If you go there make sure you go to a shooting range and shot an AK47. An experience you will never forget. If you need other info feel free to email me.

Yes, the best way is the plane!

The cheapest way is by bus.
But it's only for the strong minded!
From the borderline to Siem Reap (Angkor Wat town) it's anywhere from 8-14 hours depending on if any bridges have collapsed (which had when I went) and if your 'bus'/death mobile doesn't break down (which ours did) and leave you walking for an hour or so in the middle of nowhere haha.
It's a great experience though, but you just wont be thinking that when it's happening.
It's only 162km or so, but the road isn't paved.

If you are planning on doing a bit of travelling around Cambodia, then perhaps you could go into Cambodia by ferry (take a bus from Bangkok to the border town... forgot the name) to Sihanoukville, then you can take buses from there to either Phnom Penh or Siem Reap.

If you do prefer comfort, I would suggest the plane though.
The ride into Siem Reap was the only bad bus ride we had, the rest of the roads were fine, and the buses actually had seats and air con. Just that inital ride haha.
Bus from Bangkok to ferry, I cant remember the price... ferry was $15, buses through Cambodia are dependant on how they are feeling. Usually around $6-$12.

Taking the bus from Bangkok to Siem Reap was 250 baht. (about 30 baht to $1US)

Get a flight from Bangkok to Angkor (Siem Reap) airport on Bangkok Airways, they fly 3-4 times a day.

Expect the cost to be in the region of B4000 each way.

Hey Iggy - I was promised a first class bus and given 3rd.. but it was ok right? - So, they stop for two hrs to kill time - but thats fun right? we get to Siem Riap at night and they lock the doors on the bus while we go through town - they tell us it is very dangerous - they want us to stay at their guesthouse - I like clever people!! - I have a migraine headache from the heat and dust.. When they finally stop the bus - at their guesthouse, everybody makes a run for it just to get away from these people.. I am with my wife and moving a little slower, but we are having a great time being in Cambodia.. its vacation right? - So they grab me by the shirt - surely a local custom of welcome and start yelling in my face to stay in their guesthouse.. His breath is wonderful and i consider this a possibility until he lets go of my shirt - and then I make a run for it like everyone else.. it was a great time, but next time I will probably fly..

On the bus ride back, people twenty years my junior were sick and in spasms from the bouncing on the bad roads.. I volunteered to stay behind because they were going to abandon this poor sick girl in the middle of cambodia.. after an hour she got it together.. they waited.. - well, yes, lots of fun - but next time the flight might be a better choice - the price won't matter..

take a plane to Pnon-Pen (the capital); and a tourism bus to Siem Reap (about 7 hours away...)
i took a bus from Bangkok to Siam Reap, very long, but ill never forget it... actually, did it twice....
