i have found the best way to tone your stomach is strenuous cardio vascular exercise.....so, find an exercise that you like to do & that makes you sweaty while doing it & stick to it, the stomach flab will go away
an hour 3 or 4 days a week should do it!

holding in your stomach/tightening the abs can definitely tone muscles. however, the only way to lose the big gut is to continue to diet. When you excercise or work a certain part of the body, it only works on the muscles, when your body burns fat, it doesn't do it in just the areas where the muscles are being worked, it takes it from different places, at different times. For instance, some people, weight gain goes 'straight to my thighs", and for most of them, that's the place the body takes from last to burn calories.
So to answer your question, tightening your stomach will help tone your muscles, but it won't really do much to help the fat there - that takes diet and excercise.

Someone has developed a specific routine aimed at improving your entire waistline and in particular those 'love handles'! Normal sit-ups are not particularly useful, you need something much stronger that will also improve your abs. "Twisting" is also pretty good and easy to do to music. Even some ways to relax can help - which is my idea of fun! Unfortunately there are some exercise you can do that actually make you thicker and you should watch out for! Just think the more weight you use when holding your stomach muscles the more toned it will get. Lay on the floor and do the same if you refuse to do sit-ups, but your already halfway there. Also remember, that doing that wont burn enough calories to actually see the muscles you have underneath the fat. If you really want to see results, burn more calories than you intake and try to exercise ATLEAST twenty minutes a day. So eat lots of celery and fruits, and try to burn them off as quick as possible. Most people who have overeating tendencies find it easier to exercise before you eat, not only as a goal to get to the food, but when you eat it makes you tired quicker, and after you exercise you wont want to eat as much.

Do some work with an exercise ball. About.com has a great workout program for the exercise ball. You can also sit on the ball at your desk for active sitting. I'm not sure how much active sitting helps, but it improves balance (or at least it helped me with balance). Also, if you're more off-balance, you work your core muscles more, which means better abs!
