The first one is correct if you add a word like The or An in front:
The American speaks English.
The word American can describe an adjective, like He is American and They are American.
Is it also a noun, He is an American and They are Americans.

It should be the third one. There are more than one American, plural is Americans. Speak is also plural, so it should be Americans speak English.
If you add the word "the" in front, then the correct answer will be the first one, "the American speaks English". This is because "the American" is referring to one group of people, making it singular. Speaks is singular.

You know I have been told by many people that english is THE hardest language in the world to learn!
I personally can speak English, Spanish, German and French to name the main 4 and I can honestly say that out of all of them it took me 14 years to learn english where it only took me a couple of years to learn the others.
