To have the right to live and work permanently in Canada, you need a Skilled Worker Visa. The process is pretty detailed and will probably take you 2 years to complete. You will need to be able to document your education, work history, FBI record, history of living arrangements... You will also need to prove that you have something like $10,000 CAD in your possession. (Unless you already have a job lined up.)

An American needs to have landed immigrant status in order to move to Canada to live and work. Basically, what you are requesting is immigration, and there are certain hurdles you have to go through before that status is granted. A landed immigrant is not the same as becoming a Canadian citizen, but it admits you to the country, gives you all the rights and privileges, except for voting, holding political office or working as a civil servant.
Check with the Canadian consulate for complete information.
