The Shiite believe that the successor of Mohammed should have been Ali but he was deprived of his right by the Sunnis. The Shiites also celebrate the martyrdom of Hassan and Hussein, which Sunnis do not recognize, and who are also direct descendants of Ali.
In practice, there are few differences between the two Muslim Sects. They do not have any books they differ on, which are material to their practice. The Koran is widely held by both as the Word of God or Allah. They pray a little differently. The differences in their religious practices are smaller than those between Catholics and Protestans, for example.
The disagreements between them are more often Geo-Political.

well, after Mohammed died, everyone was wondering who should rule over the Muslim people, and the religion. Basically, there were two factions. One faction, which came to be known as the shiites basically wanted Mohammed's son, Ali, to be the next caliph. The Shiites believed that only descendants and direct relations of Mohammed were worthy of being rulers. However, the Sunni branch believed that anyone could be the caliph as long as he showed merit
