If you run for more than 20 minutes a day and eat less calories than you burn, yes, definitely.

you can't really lose your butt, unless you sit on it all day. Then it goes flat. But you can tighten it. Lunges are the best. Better than squats even. Try forward lunges and back lunges (where you take a large step back). It works a different muscle. Good Luck.

With any form of cardio you will lose weight all over.
