Without needing to go get a trap.

Its because the cat at the local barn had another litter of kittens(i think there may only be on ethis time) and if I get a job I am going to take the kittne(when its old enouhg) and the mom to get there shots and get fixed. The mom is used to humans and we could catch her but the kitten is to fast.

The kitten is in the hay loft so I cant block off the stairs and it has all kinds of holes in the hay to run into

You can use a long handled fish net, but you have to be close enough to the kitten to catch it.
The easiest way is to buy or borrow a box trap. Use tuna fish or canned catfood for bait.
Start feeding the cat and kittens every day in a corner of the barn. Call "here kitty kitty," each time you set the food out. This lets the kittens know that food is coming. Place the food near the box trap. After a couple of days, put half of the food just inside the boxtrap, and the rest of it in the very back of the boxtrap.
When the kitten is caught, remove it in a small room because it can run out of the trap before you can grab it. Also wear heavy leather gloves so you won't be bitten or scratched.

I save old food scraps that our family has eaten (or we would otherwise throw away) and put it out...then crouch down by the food. They are usually pretty skittish at first, but if you just sit there and don't move while they eat, they let their guard down. Make sure you wear gloves, because kittens will scratch and bite if they feel threatened (they don't know what's happening to them--for all they know you're trying to kill them!). Gently reach out and pet the kitten, but don't attempt to pick it up. It may shy away a few times, but keep being patient. Finally, once the kitten is accustomed to you, go down like you're going to pet it, but instead grab the scruff of the neck firmly and reach with your other hand to grasp the front feet. Pull the kitten into your chest--it will make it feel safer and not struggle as much. Then make sure you have a pet carrier close by so you can gently push the kitten in there. Let it sit there and get its bearings for a little bit--sometimes a little food is good but they usually spill the water. I would keep the kitten in an enclosed area for awhile--until you know you can catch it again. Make sure you always make your experiences with the kitten positive so he/she will grow to love you! :)

You need to define "best way." Are you primarily interested in getting the cat out of the barn forever? Or is this s family pet you want to bring back into the house?

Are you oppossed to every kind of trap? A l;arge net set under a dish of food/milk could be pulled up to catch him...

Place some cat food/milk out in th ebarn, and lace it with a medication that will put the cat to sleep.

If you could put some food in the corner or something, and get some people to help circle around her. Plus close off any rooms or lofts or whatever, so that you've got her in a more contained area.

Set out some kitty food, then check up on it alot. Once it gets the idea that it will get food there often, it should let it's guard down some. Then, sneak up on it and catch it (probably still a good idea to wear gloves so you don't get all scratched up).

Is kitten still nursing? If so holding mom is key. if not withhold food for day (or so) then bring in food. Hungry cat's come running and often don't even mind if you are still near. It won't hurt them to be without for a day.

#1- put out kitten food and wait for it to eat it then using a blanket or a net catch it!

#2- Get a string or a toy or something to catch it's eye and lure it outside to you and catch it then!

Milk and a cage traps to pen them into. Not harmful but helpful to getting kittens.

go grab a can of tuna, and take it into the barn and open it, the cat will hear that sound and then smell the tuna and come running to you to get some of it. I have tried it before and it works everytime.

With food... Everything can be manipulated and corralled when its basic instinctual needs are being met.
