Any time in the gym is better than nothing. Even if all you can do on a given day is a walk around the neighborhood for 15 minutes, DO IT!!! If you're looking to lose weight, do any type of cardio or circuit weight training with light weights. You should NEVER spend more than an hour working out. Anything more than that and you risk burning muscle instead of fat. Not good.
In terms of food, try to eat more of the following: Nuts, beans/legumes, green veggies, lowfat dairy products, eggs (high in protein and "good" fat), olive oil (also high in "good" fat), lean meats (especially fish and poultry), whole grain breads/cereals/pasta, and fresh fruit (especially any type of berries). Also, chocolate is good for you, but NOT in quantity, so have a little bit here and there -- you can have a little bit every day if you like.
Here's a list of stuff to avoid: Refined sugar and/or flour, high fructose corn syrup (found in soft drinks and just about every kind of junk food), trans fats (partially or fully hydrogenated oils), and try to keep saturated fat to a minimum.
One last thing: Reward yourself at least once a week (1/2 a day a week is better) by letting yourself eat whatever you want, but get into the habit of eating until you're satisfied. Don't stuff.

The answer to this question is actually quite long. I think the best way for you to answer your own questions is to do some research. Head on over to the book store and pick up a Women's Health, or Muscle and Fitness Hers, or Shape or Fitness magazine. They are filled with solid workout plans and diet plans.
The short answer to your question is to hit a variety of machines -- especially if you are new. Figure out what you like to do and what works for you. If you are bored on a certain machine, move to another. Just become comfortable in the gym and get used to working out.
To lose weight you basically have to consume less calories than you burn. This takes some knowledge and you should use a website such as http://www.fitday.com. It's free and helps you monitor your activites and makes you accountability for what you put into your body.
My general rule of thumb for healthy eating is to keep it low fat, low refined sugar, and moderate amounts of protein. Some fats are okay (olive oil, etc). I don't eliminate much from my diet, but everything should be in moderation. Again, knowledge is power here and those aforementioned magazines will help you establish a healthy diet.

Ideally, you want to keep your target heart rate at a consistant level for at least 30 minutes. This can be acheived by doing either weight lifting or cardio exercises.
There is no such thing as losing weight in a specific area. You have to view your fat as a whole, not specific locations. Example: Doing a million sit ups a day doesn't gurantee you washboard abs.
The rule I follow when eating is, the less processed the better. Stay away from fried food, anything high in sugar and sodium. If it has a drive-thru... stay away. What you want to eat is a lot of grilled chicken, fish, vegetables, and grains high in fiber. Also, make sure to drink a lot of water.

Ask at your gym what machines to use, and I think an hour each trip should work. As for what not to eat, you should be able to eat most everything if you are working out as long as you eat in moderation. As to what to eat, try eating lots of fesh fruit and veggies as snacks.
