There are tests now that detect the pregnancy horomone the day of your expected period. Being that you are 45 I would say yes this is naturally a time where menopause is likely to begin. The best bet is to see a OBGYN.

Most tests claim to come right, after the first days of missed period. It actually measures the amount of hormone that comes with your urine, when you are pergnent. That hormone varies for everyone. For the best possible answer take the test at you first urine begining of the day. As you rest at night, the hormone accumulates and the test strip gets enough to give you a clear result.
It sould work for any given time, as the hormone should be present if you are pregnent at all times, only that the quantity may be more as you are rested and have not possibly urinated with in the past 8 hours.
Hope this info helps.
From my personal experience, when I did at no particular time when I was asuming I might be pregnent, the lines where very dull and hence confusing. To make myself sure I did the second test in the morning and the lines where much readable and I was sure.

Some tests are more sensitive to the HcG hormone than others (http://www.fertilityplus.org/faq/hpt.html shows a list of most tests in the order of their sensitivity).

The hormone is not detectable until the zygote attaches to the uterine wall. Once it finally implants, hcg is produced and steadily increases (I believe the numbers double each day), reaching its peak 2-4 months into the pregnancy (I believe it peaks at 4 months, but i could be wrong). A blood test will give you the fastest result, if you are testing early on.

I'd recommend making an appt to see your ob/gyn. He or she will be able to let you know what is going on.

Good luck!

Some times it takeslong time for the ladies to get their mensuration because of various reason and with one i can help you is might be you should be in too much of stress and for that affects the routine system of the health.
Even you got negative reasults on the home test the reason should be what i told you earlier or better you consult a doctor before it's too late.

Within about 5 days after your first missed period.

Most home tests can give you an accurate result within days of your expected period.

i think its something like a week after your missed period. but remember, false negatives are fairly common, so try it again. if you still get the same answer, see a doctor and just get some blood work done, they can tell you either way very quickly
