Whether removal of dat file will affect the functioning of windows?

yes, but sometimes they are hard to delete, especially "index.dat". To do so, reboot your computer, and when the manufacturer's name comes up (Compaq, HP, etc), hit F8 repeatedly. You'll see a screen of boot choices; select "safe mode". Once Windows boots into safe mode, search for the .dat files and delete.

It depends what that .dat file was ascosited with. It usualy means a incomplete data file and usualy will not make windows unstable. So it should be safe to delete.
