I would think hamburgers in america. Hamburgers of america sounds like the name of a fast food joint.

One. The hamburgers are not inherently from America. Hamburgers in America are super-sized is the most accurate.

Has to be two...

If one was to bring a tiny hamburger from Japan to America it would suddenly be big or 1 would be false.

Hamburgers of America conveys the origin within the borders and if they are big it would be consistent always.

It depends if you want to emphasize the place or the actual hamburgers. I'd say the first one sounds better though.
