Japanese Sumo wrestlers face problem while travelling in a commercial airliner...so sometimes they charter flights with bigger seats!

Except the above...a person with 250 pounds weight will have problem during the long haul flight in the economy zone, as he would not be able to sit propoerly. However, the first class & business class have bigger reclining seats which offer better comfort.

The major problem is that such persons will face more challenging situations during an emergency landings or any crash in accessing the emergency slides.

Nothing else!

Sure they do. I'm personally not overweight or have a big bum either, but I sure saw some fat people having difficulty walking through those narrow aisles! They have to buy a first-class ticket, or two economy-class tickets to sit down properly!!!!!
