In ACTS Chapter 2 is the Answer. It was on the Day of Pentecost, 50 days after the Lord was crucified.

By first church, I assume you mean in the Christian set-up.
Read the book of Acts (of the Apostles).

I think that in B.C. times, people would just start to meet up a certain day, and then they just made it a permanent thing. Pretty cool.
