Basically if it's a two class flight there is no real difference. On a 3 class flight i.e. 1st, Business & Coach is where you really see it. Obviously, because these are bigger planes they are almost exclusively flown on longer routes i.e. cross country or over oceans. It depends on the airline really. American calls their first premium and I have to say it is awesome. Nowadays business is always full of rude business people using upgrades and taking all available space for their laptops, newspapers, etc... A continuation of our me first society enclosed in a tiny tube hurtling hundreds of miles per hour in the sky. Times I've been in Premium have been great. It is hardly ever full, I've been the only one up there on many occasions. Usually caviar prepared at your seat, better quality meals, food bar open for entire trip at front of cabin, restrooms for fewer people, individual TVs for your seat with choice of movies for each person, first on/off priviledges, first luggage receipt and my favorite - fully flat reclining seats to a bed position. Drawback - you pay dearly for it. Unless the company pays for it I would never spend the money for it.

First class is the ultimate luxuries and nice comfy seating. Business class is close, but it is geared towards business travelers, so more techno-friendly things, less luxury.

First Class has better amenity kit, less seats, more leg room, better food, more expensive alcohol, and seats that can be fully flat for sleep.

first class is basically just better quality, when i flew first class, we deffinately got better meals, i dont know if its still the same though. basically you have more comfortable chairs and maybe a better view of the tv, its not too amazing.
