So many choices.

"This is she" is the correct, formal response.

You could also say, "This is (your name)." That is very nice.

"Speaking" is another fine answer.

"It's me" and "that's me" are informal answers.

Sorry, "This is me" is incorrect.

You can ask, "who is calling?" before revealing who you are to the caller. This is usually a good idea.

Rude answers include, "Why?" and "What do you want?"

The correct answer is "This is she." Because me is an objective case pronoun meaning it must be the object of a prepositional phrase, or the direct or indirect object of the sentence. In this case it is a nominate case pronoun explaining who is on the phone.

This is she is correct english, but if anyone gives you a hard time for saying it the other way you can go ahead and kick them in the teeth. Tell them it's from me.

Well, I read about this in the dictionary a few days ago, oddly enough. You can say:

It's me OR It is I.

When using a contraction, "I" becomes "me". Weird, huh? Also, "This is she" would be correct.

this is she, I think. Because if they ask for Jamie, but you are Cindy then saying this is me is stating that you are yourself, Cindy not Jamie. But if you say this is she they you are agreeing that you are in fact the person they are looking for.
