If you are truely a friend of this guy it would not matter what his sexual preferences is. I truely believe that waht a person does in thier bedroom and with who is no one else business. Anyway to answer your question I don't think you can be sure if this guy may be gay. Only the individual will know for sure and it is up to him to tell others. i don't think the lack of a girlfriend throughout this guys life is a sign he is gay, maybe he just isn't interested in the whole dating scene. You just straight out ask him and hope he's honest with you.
And not having a girlfriend doesn't make him automatically gay. Maybe social awkward, or very picky, but not automatically gay.

If he knows styles of women clothing or talks like a girl he is gay. A guy who doesn't look at women but instead men he is gay. If you see him working in a spa and salon place, well you know what he is.
