"is", because the subject of the sentence is man. Man is singular so the verb tense is singular. the prase "and two women on the bus" is a conjunction, and not the subject. Your question is a subject verb agreement question.

Other wise it is: "There are a man and two women on the bus."
If you take out the conjuction "and two women on the bus," you have:

"There are a man."
which is not correct.

"is" cause it is refer to the first word after it "a man" and not refer to the whole " a man and two women"
