Go out with her.
Maybe if you think there might be tension between you and her brother then tell him of your feelings for each other. Kind of old fashion but it does show a respect that may lead to a positive response if handled the right way then everybody is happy.
Are they fraternal or paternal twins? Maybe it could be taken as having homosexual feelings for him.

So what? Just go out with her! It's even better, you can ask for help, I mean he's your best friend right? But if he's kinda overprotective over his sister, you should ask permission from him, but do it slowly. Start by showing her a little attention and so on, your best friend is bound to realize it some time later on; and that's when you ask him. But do it slow, not too shocking or anything.
And if you do hook up with her, don't start telling every little detail to your friend, he might not like it and get a little jealous, he might think that you're stealing his sister away from him(if they're really close).

I think you should talk to your best friend about your feelings for his twin sister.. Not that you're asking for permission to go out with his sister - that you don't have to do. It's just a sign of courtesy and respect to him as the brother.. ^_^

You should still be able to go out with her, but keep those relationships separate. As you might with a different buddy, don't talk about your dates (with his sister) with him, or brag about what y'all did.
Realize also, when the relationship between you and she has run its course, there may be issues if she's feeling hurt, her brother may feel the need to or console her, and may have hard feelings toward you for a time.
Make sure your friend knows that you're going to ask his sister out...keep everything on the up and up.

Tell your friend your intentions first. Don't try to hide your attraction for his sister from him. Things could all go well. My aunt ended up married to my uncle's (her brother's) best friend. She is still in love with him to this day, over 20 years after his untimely death.
