Sense of humor definitely with a good smile that comes from the eyes too. She has to be intelligent too and have depth. By depth I mean she has opinions and thoughts on deep subjects not just superficial stuff.
Physical looks are important, but every guy likes something different, so don't worry about that too much....every girl has something about her look that some guy is gonna love, guaranteed.
