the most powerful one i know of is a custom handgun that fires a .50 BMG round, it comes with a warning not to shoot it course of the risk of wrist injuries.
the current most powerful production handgun is the .500 mag.

I guess you would have to describe how you define powerful. If you are talking about foot pounds of energy at the muzzle it would be different than foot-pounds of energy at say, 500 yards. While most handguns are incapable of delivering a projectile at 500 yards, some are.

Then you have to allow for multiple shot capability. A fully automatic .22 caliber weapon with a 50 round magazine could deliver more foot-pounds of energy than a .44 magnum revolver.

There are actually handguns produced to fire practically every cartridge made although I would not want to fire some of them.
