You need an editing program, source footage (best to use DVD, you'll need a ripper for that), and music. I make AMVs and it's alot of fun. There are some wonderful guides as well as links to all the stuff you'll need at animemusicvideos.org
Here's what my process looks like
1. DVD decrypter then I make a lossless compressed avi in DGIndex.
2. Use AviSynth and VirtualDubMod to clean up (deinterlace is the most important) footage.
3. Use Exact Audio Copy to make an uncompressed .wav from CD.
4. Start editing! Right now I'm using Premiere Elements, I kinda like Ulead Video Studio 8? better because it had more effects.
5.Render it lossless! Separate audio and video.
6. Put your audio from your project into BeSweet and compress it to mp3.
7. Put your new mp3 and your project video into VirtualDubMod and compress it.
Voila! You know have an excellent looking and sounding *small filed* video! The small file+quality is the reason for all the hullabaloo. But read the guides there, they tell you all the codecs you need and whatnot. Good luck! and Have Fun!

...added a link to my profile too just so you know I'm not lying when I say I've made these before. :P I only read the guides before doing the last 2 and you can totally see the difference in quality and size. I'm never going back to the way I used to make them which was slightly easier but just not worth it. ^_^
