Use a level. Mark it at 12" and set it on the roof with the bubble at level. Measure from the roof up to the 12" level mark at 90 degrees. My roof is 12-3 pitch. That is for every 12" it drops 3"

buy a pitch and angle finder for about $10. just lay it on the roof and it will tell you the pitch. Finding it with a speed square takes too long.

The pitch on a roof is usualy determined by percentage of drop.
ie. a "four to one" is four feet out with one foot of drop at four feet.
