Just ask her about her boyfriend. The best conversation start out asking about the person being approached. Say "Hi! I am Randy Wolff, and I couldn't help but notice that necklace, it is so pretty where did you get it?" Followed by "does your boyfriend like it too?"

You cannot really tell unless you ask her point blank to her face. (alone or quitely would be better) Of course you could just go over to her and start talking and toss in some topics about GLBQ issues, see how she reacts. Get close to her and see her reaction, but if she is shy that could backfire. Ask her friends. The only true way is to ask or ask her out. She could be a feminine lesbian, bi girl, or straight - just have to jump in there.
As for pick up lines, I'm going to go with saying something nice about her look. Like telling her she has nice legs. Gets right to the point.
Edit: I like Randy's question about asking her about her boyfriend too, but that won't work with a bisexual girl half the time.

Just ask her out, the worst she can do is say that she doesn't go that way. Then you'd know. That's what I have always done, ya, it can be awkward sometimes. But I think it's better than all the hinting and innuendo and beating around the bush! Just get it out there and get it over with. As for the best line? It's really cheesy I know, but it has always worked for me. Look at her till she makes eye contact with you, then walk over to her and say "excuse me, I don't want to interrupt but I couldn't help coming over here to you, your gorgeous(insert color) eyes just burned a hole right through my heart and I couldn't walk away. Please forgive me if I have intruded, can I buy you a drink?"

Come out and tell her your a lesbian. If she still wants to hang out with you that's great. Next invite her somewhere that takes her out of her element and into yours. If she still wants to hang out with you, it's cause she is interested.
Best pick up line: Pretend to wipe something from her face, and look her in the eyes and say 'perfect'.

The honest thing to do would be to just ask. Many Gay guys do that and believe me, it's much appreciated from us straight guys rather than creating an potential embarrassing situation for both.
