The Classic American Southern accent is generally believed to have developed from the 17th century English "West Country" (the south western part of England from Bristol down to Plymouth) accent. Here is Answers.com on Southern American English:
Southern dialects substantially originated from immigrants from the British Isles who moved to the South in the 17th and 18th centuries. The South known for being largely settled by English from the West Midlands - the West Country. (The West Country dialect of Britain is also very similar to the Southern dialects.)

Many authorities believe that the accents of the South are more directly linked to the English spoken during the 17th century. Jamestown and all that. The New England Puritans had spent some time in the Netherlands, and their accents would have been altered slightly. New York was also settled by the Dutch, who would have lent a further dimension to the language. Other northern influences woul;d have been the Germans and Scandinavians, while in the South the Scots and Scots-Irish settled in the back country.

Just a wild guess, a stab in the dark if you will, but I will say the Southern States, say from about Kentucky down to about Alabama. It kinda stops at Florida, much like the inbreeding.
