It depends on the car. Scions can't come off sticker at all, but that's ok, because resale is very high on them. Newer, hotter vehicles typically cannot be had with any discounts, and if there are any, they will be small.
you don't want to shop around for the biggest discount, because that isn't neccessarily the best deal. The best deal is the car of your choice, with the options/color you want, at a fair price.
Check: http://www.edmunds.com to see the differences between invoice and retail. They will also tell you if there is any dealer money on the car. Dealer money is like rebate, but it goes directly to the dealer, and they don't ususally mention that to you.

If a dealer wants rid of a vehicle, they will go as low as what they paid for the vehicle. I just bought a 2005 Explorer for $7,700 less than sticker. It was new with 290 miles. The dealer wanted it off the lot, and the salesman was a friend.
There are end of year rebates, customer loyalty rebates, armed forces rebates, college grad. rebates, and if your lucky they will use "hold back" money. The "hold back" money, is money in the price on the sticker, for advertising and other misc. items. The key is to shop around, and find a salesman that wants to sell a lot of cars, not a salesman trying to make a lot of money!

use kelley blue book online to find out the value.
they give retail (what a dealership should charge)
and wholesale (what you can trade in for)
and private party (what you should get selling it to another individual.
you also put in the condition etc. most everyone uses these figures to start off with the price of a car.
Car dealerships are "allowed" to come down as much as they want, though your particular salesperson will not be allowed to come down more than a certain amount. If they come down too far they will lose money on the deal.

When they say "No" to your offer. Then go to a different dealer. The managers make the final decision.
