If it is truly your best mate then instead of calling his missus an offensive name give him proof. Prove to him that she is cheating on him, that she uses crack and that she is a whore. If he cant see she is nasty, take him to get some eye glasses. If you live in a free country then there is nothing wrong with stating your opinion. I just hope that your best mate can appreciate your freedom.

It's not subtle though. Maybe add a little subtlety? Does she have any good points? Try hard, strain....mmmnnngggg.... did you get any? If so, think of those and tone down the nasty cheating crack whore gag a little.
Meanwhile, some deconstruction:
"cheating" and "whore" are the same thing in your book, I'm guessing. Slavery outlawed, it's her sex life. Whether she's playing fair with your friend is a matter of judgement, and a very thorny one at that.
"nasty" is subjective - you might be right, you might not.
"crack" - really?

Well first thing is first do you know for sure that she has done things that would hurt your friend? And have you told this friend and given him proof? If so then yes you have every right to, especially if you are trying to make him realize that he doesn't need to be with her. Unless he has forgiven her then the answer would have to be no. If he knows all the facts and says he still loves her and forgave her then being his friend you should support him and his wishes. You don't have to like her but it is always important to give support to a friend, or you will end up being the one on the outside looking in. Just be there and hope that down the line he will see what everyone else does, but if she is call her on it. There is never just one answer to these things just what you think is best, follow your heart and you will see that everything always comes out on top .

Just because it's true doesn't make it nice. You have the right to say absolutely anything you wish, but they also have the right to be offended. You just have to decide whether having your friend like you is worth pretending his missus isn't a nasting cheating crack whore.

You can still be accused of slander even if what you say can be proved to be true. This is often not appreciated. The test that the law applies, is whether you intended to publically defame her character. In any event, the description you state above appears to highly subjective, so it might be very difficult to prove anyway.
