it depends on whether you take it at the first sign of the sore or after it has opened, if early it may go away in 2 to 3 days(still complete your course of therapy) if you waited until it opened it may take up to a week(again take the whole course of therapy)

Acyclovir will probably make your cold sores heal a bit faster, than without it, if taken orally. The main purpose of Acyclovir, is to make your outbreaks, much less frequent, and in some cases cold sore breakouts disappear completely. Sometimes cold sores do disappear completely, with a breakout occurring, only once or twice per year. When you do get a breakout using the Acyclovir, apply Zovirax topically, at the time as you take it orally. Taking Acyclovir orally and applying it topically during breakouts, works best on getting rid of active cold sores.

If you are using the cream then you should FEEL relief soon. Otherwise it takes 3-5 days for it to go away. Pills take the same amount of time but don't work quite as fast on the feel good part.
