You burn more calories if you increase the incline because your body has to work harder (assuming you keep it at a reasonably fast speed).
Usually the way to go is to do 2 mins at 0, 4 mins 2%, 4 mins 4%, 4 mins 6% etc until you're at 10% or higher and then stay at the highest incline for as long as you can without decreasing the speed, then slowly decrease the incline 1 min per 2% to bring the heart rate down.
