No, not unless you are deficient in B12. You only need a few micrograms a day to meet your B12 quotas. Most Americans get plenty of B12 through their daily consumption of meat / dairy products. Getting a shot of B12 would probably provide a placebo affect but it certainly wouldn't lead to weight loss and this is certainly the first I've heard of it.

Remember weight loss is achieved by consuming less calories than you burn. For example: burn 2500 calories a day through living / exercise and eat only 2000 will eventually lead to weight loss.
