Quite coincidentally, I'll be going to NY for a week with my lovely wife as well next week!
You're not really missing much not having a car. In fact, driving a car in the Big Apple is just an extra hassle to deal with. As a result, I don't intend to drive in NY.
NY has one of the largest subway systems in the world; I would suggest you do as most New Yorkers have been doing everyday: take the subway. Take a taxi, if needed. & most importantly, wear a good pair of sneaker, you'll do a lot of walking in NY. Blend in. Be like one of the New Yorkers for a week. I bet you'll like it.
Since you're there, check out: the World Trade Center site; Radio City Music Hall; view NY from the top of the Empire St. Bldg.; Financial Dist.; Theater Dist. (catch a broadway show if you wish); Rocketfeller Ctr.; St. Patrick Cathedral; Statue of Liberty; NY Chinatown; Macy's Shopping Ctr.; Times Sqr.; Penn. Stn.; Madison Sqr. Garden; the United Nations Bldg.... and many more....
If you'd stop by any Tourists Info. desks located by any of the sites mentioned above, ask for a tourists info. booklet, you'll find even more interesting destinations to visit.

Everyone has pretty much said it. You can buy an unlimited weekly metrocard and get around everywhere you need to (even the airport if you don't care about lugging you bag)
Here are my favorite must sees in NYC:
Times Square
Empire State Building
Central Park
Greenwich Village (walk through)
Chinatown (get your trinkets on Canal St. - should haggle over prices with street vendors)
Museum of Natural History
Coney Island and ride the Cyclone
Yankee Stadium or Shea (Let's Go Mets!, #1) and see a game
Ride the Screamer
Skip the Circle Line, it gets boring. You can ride the bus that goes down 5th Avenue and see more exciting things.
Nightlife you can find great places all night! The city that never sleeps!

Go to Times Square, 42nd street and Broadway.... Definitely go on the Circle Line - boat that goes around the entire island of Manhattan. It goes right by the Statue of Liberty and there are amazing views of NYC skyline from the harbor. Also definitely go to the top of the Empire State Building. get there early, the lines are long on weekends. Canal street shopping is a good experience. Go downtown and check out Ground Zero -it's very moving, and while you're down there, go shopping at Century 21, it's a zoo most of the time, but designer discounts, huge store, amazing NY shopping experience. Have fun, and don't mind the locals, we rush things, but we're not that bad... just in a hurry.

definateley go to the top of the empire state building, better if you go at night, a clear night. The view is spectacular during the day, but at night it is truly amazing with all the lights--pick a clear night though.

lombardis pizza, 32 spring street,in soho/little italy the best authentic italian pizza in new york, its sit down only, you have to order a whole pie not by the slice.. its right in the middle between soho and litlte italy so u can explore those great neighborhoods while youre there.

the best way to get around is by subway and /or bus, if youre here a week, buy a weekly metrocard at any subway station. its 24 dollars per person, you can ride the subway and buses as much as you want for 7 days for that one price.
