Say "Do you have a girlfriend?" I would appreciate that sort of directness. Plus, it implies that you're interested.
If you want an alternative...
I say something like "Did you spend the weekend with your girlfriend?" Then he'd say "What? I don't have a girlfreind." and then you say "Rrrrrreeeeeaaaaalllllyyyyyy? Interesting. Why not?" Then he'll say "I just haven't found the right one." And then you're in.

how does it matter that he has a sister?? anyways...if he knows that you like him then he'll probably tell you if he has a girlfriend or not...you can get it out from his friends or someone or simply ask him directly...
