why must the spot be above the solvent level during Thin layer chromatography???

So that it doesn't separate out into the solvent. The goal of TLC is to have the solvent carry the "spot" up the plate, separating it, so that you can measure the different components and identify them. If it starts off in the solvent, it all separates into the liquid, and not on the plate.
