They are the same vehicle just two different engine sizes and yes price points. IS 250 it is a zippy car and plenty fast. 0-60 is 7.9 seconds. The IS 350 is 5.3 seconds. The IS 350 has a more advance stability and traction control system (VDIM vs. VSC) to handle to increased power.

And of course with a smaller engine you get better gas mileage. IS 350 (21/28), IS 250 (24/32).

The IS 350 is only available in rear-wheel drive. The IS 250 has choices of rear-wheel drive, all-wheel drive or 6-speed manual transmission.

Some minor differences you can check on Lexus.com, but main one is the IS250 you can get with all-wheel drive, but IS350 you can not, only rear-wheel drive.
