Probably the discovery of DNA. Although it may not seem like it yet, it will eventually revolutionize the practice of medicine. In the future, doctors will be able to identify and treat disease more effectively through genetic manipulation.

Depends on whether you consider applications. Quantum mechanics has a lot of applications, but it's not really "beautiful", while general relativity doesn't have many applications but it's intimately connected to geometry and is a beautiful theory.
