I'm a firm believer of not touching your friends sister but theres always exceptions. Ask him if it'd be cool and only do it if your intentions are pure. If you're at fault if the relationship goes sour, you'll most likely lose the girl but a friend too.

I think that if you are best friends with her brother then you should be good enough for his sister. Unless he knows you well enough to know that you aren't the kind of guy that dates girls with good intentions. If that's the case, do your friendship a favor and just pass this girl up. If not, there is no reason why not to explore your feelings for this girl.
