First of all, you have to figure out how you'll use the bike. That will help you pick the right bike for the right purpose. Then you need to figure out a budget (remember to factor stuff like a helmet, gloves, and any other gear you feel you need).
Once you've done all that, start doing some test rides at the local bike shops. Figure out what you get for your money (free service, discounts on accessories, etc.), and make sure they take the time to find a bike that fits you correctly - how a bike should fit you varies by bike type...but you should not be able to sit on the seat and have your feet flat on the ground like another response mentioned. That's falt-out wrong.
If a salesperson tries sticking you with a wrong size to clear out the last of the inventory, find a different shop! Fit is everything.
A lot of other buying advice I can give you is specific to the type of bike/riding you'll be doing.

Depends on how you want to use it. Off Roading? Race? Liesure?
You need to pick a bycicle that fits your purpose. Then you need to know the style for that purpose. You have bikes w/shocks and springs on the forks, bikes w/shocks and springs on the frame itself. You have trick bikes, bikes w/ major gear ratio.
I really depends on your style of enjoyment or aggression.

Gp to a good bike shop where they know their stuff and they will tell you all you need to know. Trust them as you will be going back to have it adjusted, new bits fitted, a service etc etc.
Best thing is to have a syle of riding you want to do and tell them a price you will go too and let them advise you. Real bikers like to share and will be really helpful, just dont get all the advice and buy it cheaper from a mass cycle depot and then expect them to service it when the back wheel falls off!!

FIT. It is crucial the the frame be the right size, the seat the right height, and the handlebars be the correct distance away from you in order to avoid injury to the knees and to ride safely.

that when you sit on the seat, your toes touch the floor. that the brakes work before you start riding. that the bike is comfortable for you (especially the seat).
