No, they're not always on corners, but they like to be. Since Starbucks is completely corporately owned, the company hires people to scout out the most visible and accessible spots for their stores. Corners are lucrative places because they can be seen from different directions, they often offer a larger property, and plenty of opportunities for there to be windows in the stores (if you've been in a Starbucks, you'll notice that they're a bit window-happy).
If they're not on a street corner, they're likely to be on the end of a strip mall. Others have mentioned that they've seen them in the middle of strip malls, and they can be. The busiest store in Wisconsin, actually, is in the middle of a strip mall.
Regarding the Starbucks in Barnes and Nobles, Borders, Targets, Jewels, etc. - be careful. Those are not 100% true Starbucks. Those are called licensed trade stores and are not operated by the Starbucks corporation. They are actually owned and staffed by the company that contains them (i.e., Target, Jewel, etc.). They pay to use Starbucks products, logos, and recipies, though they may not always follow all of the company standards 10000%. (So, if you're trying to prove your girlfriend wrong, this may or may not help you.)

Our local starbucks is in the center of the shopping center and I know another one that stands alone in the middle of the block - but the vast majority are on the corner.

No. Starbucks is NOT on every corner and if your girlfriend is arguing that then she is being ignorant. Tell her to visit other cities for Starbucks and then ask her is she still thinks Starbucks are on every corner.

My favorite Starbucks locale is in Target! I can shop the best box store in the universe with my venti non-fat latte. I LOVE Starbucks; however, I do try to frequent independent coffee-houses as much as possible.

Heck no. Where I live, I don't know of more than a couple on a corner. Most are in strip malls and shopping malls, or grocery stores and the like.

Where I live they are building one that stands alone. For example, it is its own building in the middle of a parking lot. Not stuck between other stores, or on a corner of a shopping center.

No they are not all in corners. Here in Washington State they are in stores or in malls and actually all over the place, but not all are in corners.
