No, you want "in a car accident". With "by" you'd need something like "a careless driver"--some kind of person.

"After the graduation" is ok if you've already referred to the graduation as an important event for yourself and him. (For example "he was a guest speaker at my sister's graduation--less than a week after the graduation, he was killed.") If it's his own graduation, use "his"; if he was in your class, use "our". Better still would be "less than a week after graduating, he was killed."

I keep automatically changing "one week" to "a week". "One week" is not wrong, but "a week" means the same and sounds more natural.

So I'd write this:
Less than a week after graduating, he was killed in a car accident.

you should change the word 'the' to 'his' if it was his graduation and 'by' to 'in' then it will be correct. as in
Less than a week after his graduation, he was killed in a car accident.

Drop the comma. It's the only thing that is grammatically wrong with the sentence - you don't need it; however, it would sound better if it was written like this:

He was killed in a car accident less than one week after the graduation.

i think that it'll be better stylistically if u dispensed with "the" and used "in" instead of"by".
the sentence will be
"less than one week after graduation,he was killed in a car accident."
