A joke is usually a tell-tale incident or a story that reveals stupidity, dumbness, naivety, ignorance , weakness, etc .. and we, as listeners feel superior to it's subject and the people involved in the joke, so we feel a mixture of pride, self-satisfaction, superiority, victory and -more important -a relief that it's not us whose stupidity is revealed. This blend of feelings is then transformed into a neurological language (electromagnetic current) that sends the message as is to the brain which in its turn works on the voice, facial expression and the like to expose a mimic of the blend of feelings in neurological language - laughter.

its a learned thing that we laugh when someone make a good joke. Our brain is conditioned that whenever it recieves a sound from the ears that transmits good joke..or something like that...the brain signals back to the neurons to respond to that joke and laugh

We've been conditioned since infancy to have that reaction. Infants mimic the facial expressions of whoever they are surrounded by, so smiling becomes associated with happiness
