Crazy.

According to "Dirty Harry", played by Clint Eastwood, it is the biggest hand gun made, capable of blowing ones head clean off.

To be used on punks that feel lucky.

This is where he would say, "Well punk, do you?"

Or maybe it was a .44 Magnum.

I stand corrected, it was a .44 Magnum.

It could also be a reference to a vinyl record. Less than 30 years ago, music was produced on vinyl albums and the numbers associated with them (33, 45 and 78 I think) were the speeds at which the player would spin them.

So "I've got my 45 on" could mean listening to an album. The rest of the lyrics to the song might help determine whether the singer was a just a music lover or a g-unit.

Back in the day singles were sold on 45 rpm records (usually one song per side. American Pie was so long that it took up both sides of the 45). These were used widely in jukeboxes and by radio stations. The record companies would send promotional 45s to radio stations hoping to get the song on the air.
