Broadway is doing very very will right now, and many tickets are hard to get.
My favorites are Avenue Q, Wicked, Spamalot and Jersey Boys - all hard to get tickets.
If you haven't seen any of these shows - they are worth seeing and you can probably get tickets: Chicago, Rent, Beauty & the Beast, The Lion King, The Producers. Phantom is good too... Hairspray was really fun if you can get tickets.
Shows I haven't seen but would like to: Sweeney Todd, 25th Annual Putnam County Spelling Bee, The Color Purple, Faith Healer, Drowsy Chaperone, Mama Mia and maybe Tarzan.
The TKTS booth has moved temporarily to a space just outside the Marriott Marquis. You can always try your luck there....

I would recommend two: "Spamalot" and "Jersey Boys." Since these are not newly-opened shows, you don't necessarily need to buy tickets in advance, but you may wish to do so, especially if you want popular nights (e.g., weekend nights). If you wait to buy tickets until you are in New York, you may want to check out the TKTS booth at the northern end of Times Square, as TKTS has some discounted tickets to shows (but not all shows/dates are available). TKTS lines can get long, so plan on getting there early if you choose this option. FYI, theaters are "dark" on Mondays (no shows).
