By saying that knowing is not enough, he's saying that if you have knowledge and do nothing about it, it's useless. You must apply the knowledge to some useful area. By saying that a will is not enough, he's saying that even if you have the will, it won't matter unless you do something about it. Both parts of the quote essentially are saying that you can't just be idle (with will and knowledge); you must use what you have.

Just knowing something doesn't mean anything, it's how you apply your knowledge. Willing something takes no effort you have to get up and do something. You have to be active in your life and not sit around and rot.

He means that it is not enough to have the knowledge, you must put in to good use. And you may have the greatest will and the best intentions but if you don't act on them, then they are pointless ideas.

It means action is more important than just knowing and being willing to act on something; eg., if one knows the solution to a problem and at the same time, willing to solve it, he must act on it, apply his know-how and do something...actively, not passively.
