The stairwell is in the middle of the first floor of an old building and leads to the basement. I would like to be able to remove the railing and place a cover over the rectangular stairwell (4' X 10') which would allow me to create more open space on the first floor when necessary.

I'm not sure how you could anchor a railing to make it safe and have it removable, but PVC piping comes to mind... Perhaps an anchoring piece at the base of the stairwell, then simply insert the rest of the railing into the base....

Not that would be sturdy and practical. If you don't use the basement regularly, why not remove the banister permanently and make the floor/door the norm?use a piece the right size of whatever material is on the floor around it on top of the door to blend in and you have more space all the time. Use a flush mount latch handle and you have access when you need it.

Why use a railing at all? Go minimalist- remove the railing your space will look and feel bigger. Create a hatch that pops/slides open and have it blend in with the floor material you've already got. This is probably your best bet.
