it does not have to do with drugs.
Wanton means "without motivation or purpose"
Annihilation means "total destruction"

So basically, the quote
"Nothing like a bit of wanton annihilation to brighten up the day"
means
"Nothing like a bit of purposeless total destruction to brighten up the day"

Off the top of my head I'd say wanton means, with abandon or disregard. annihilation means, utter and complete destruction. I could look it up in the dictionary and quote that for you, but why don't you do that yourself?

You look up each word by itself, then put the definitions together.

In this case "merciless destruction" fits well.
