As for a single line of code...
In December 1989, AT&T Labs patched the software on its switching network. The code was thought to be thourouly tested. Unfortunatly, a missing C break statement threw the system into error recovery subroutines which in turn took down each machine in the network. The system was down over nine hours with 74 million lost calls. It is the biggest breakdown in the US Telephone network ever.

Don't know about the most influential line of code, but probably the most famous is: printf("hello, world\n");
This is the C language statement that prints hello world in the shell or console window where you are running it. Many other languages such as C++ (cout << "hello, world";) and Java (System.out.println("hello, world"); and others have their own versions of the hello world program. This is frequently the first program a new programmer writes when they first learn the language.
