Short answer: If you're asking that question, you're not in love.
Longer answer: True love is a choice you must make every day (perhaps many times a day.) If you were just following your desires from the start, you probably never really loved them in the first place. BUT, that doesn't mean you can't! You have the power to love or not to love, but it's like a freight train: It starts slowly, but once in motion, can NOT be just stopped.
So, you have a couple questions to ask of yourself: 1. Are you going to actively love, or allow yourself to be dragged to and fro by your desires? and 2. It this person the one you are going to love?

I think when you feel indifference spending time together. When you feel it's a chore to call him and spend time with him, instead of fun or interesting or stimulating.
Love changes seasons and comes in different forms. Sometimes it's serious and sacrificial, sometimes it's fun and emotionally exhiliarating. But when it becomes stagnant, and you'd rather be alone...you can see yourself happier without the guy, then it's time to start considering leaving.
Also, if you argue a lot or have so much trouble communicating, it may be time to call it quits.
Sometimes it helps to take a break and decide what to do after some space.

It happened to me last year with a guy i'd been with for 5 years. I had to break-up with him. The signs were: I didn't feel like going out with him anymore, I started prefering going out with other people, started looking at a lot of other guys (and not just looking...), he didn't stimulate me sexually anymore, when we met we always ended up having a fight.
