You can start by saying HI, the next time you see her. If she says Hi back then continue to say hi to her and like ask her how her day is going. Then go from there. If she responds in a positive fashion or way, you can ask her if you can buy her a coffee. If she is interested in the least you'll know and you can eventully tell her. And even ask her out. Now, why are you even mentioning the Lesbian thing? THAT should have no weight in the issue at all.

Get to know her a little. Ask her on a friendship-type outing, maybe even with other coworkers.
Get a list of questions together that don't come out and say "Hey! are you gay? Can we kick it?" but test her knowledge of gay/lesbian culture and whatever's going on in the gay community right now. (been years since I've been there...no clue where to start, but if you know a good local lesbian bar, that would be a good place for you to start)
You should have a working list of ways to pick someone's mind to find out if they're interested in dating you anyway, reguardless of your orientation, so you don't end up wasting your time on someone who's not really interested.

observe her actions but be discreet. Smile at her but don't look like a maniac..Observing and learning her actions will tell you how you'd deal with her in the future. You might also try to engage a little talk with her for example you wanted to ask for time. Then be extra kind and try to engage in a small interesting talk. You'll be giving her a hint in less than a minute. Good luck!

Put your best smile on (not a freaky smile but medium grin) and walk up and introduce yourself.
You: (approach her) hi, are you a lesbian?
(smack on your face)lol
ok, just kiddin with that . Just say hello and ask her how she is. It's simple and an easy way to start a conversation. she might just start talking and you realize 'why was I so worried about?.'

What in the world would make you think she is a lesbian?? Just because she has not spoken to you?
Hey, she might think you are the biggest dork she has seen or she might think you are GAY since you have not approached her.
For heavens sake, why don't you just walk up and say, Hi my name is Tom (or whatever) and I think you are really nice, would you like to go to lunch some afternoon? If she says no...you have your answer, if she says yes...again you have your answer.

First thing would def be to start up a converstaion, just say hi, and get to know her, flirt with her after a little while and she will get the hint that you like her. Unlike men, women are good at taking hints!!

Have a get together and invite some coworkers. Tell here that you are having a party and you're inviting some people from work and she's welcome to come if she's free. Exted the invite that she may bring her boyfriend. If she's even remotely interested she will come and if she's single she'll be alone.

About anything , the only way you'll learn anything about her is to talk to her ...you'll not only find out whether she's a lesbian??do you have any indication she is one?? but you'll also find out what type of person she is , is she even single and if you guys have anything in common . Take that case of lesbian out of your mind since you don't know her,otherwise you'll turn off your feelings.
It is not a big deal if you work together,what you have to do is ask her of something related to your work,and that's when you'll introduce yourself to each other.use that little chance you'll get.
