If you have xp loaded on a hard drive using ntfs and then you install Microsoft Virtual PC - then install ME as a guest OS in Virtual PC - then yes you can have it work. BUT, as a standalone operating system, no, it will not work. Windows ME by default would be able to use a FAT16 or FAT32 drive, but not NTFS.
In Windows 2000 and XP FAT16 and FAT32 are provided for legacy and upgrade compatibility, because they were the standard file systems for Windows 95/98/Millenium

NTFS stands for - New Technology File System designed for Windows NT, 2000, XP.
NTFS is a high-performance and self-healing file system proprietary to Windows XP 2000 NT, which supports file-level security, compression and auditing. It also supports large volumes and powerful storage solution such as RAID.
And Windows Millenium Edition is not NT based, So it wont work on NTFS partition niether it will show u any NTFS partition if u have any. Windows ME will work only on FAT32.

No you would have to either reformat to FAT32, or resize and create a second partition in FAT32. You can use the following freeware to resize then format the second partition to FAT32.
